As per my understanding, SQL is not supporting to Upgrade from SQL 2000 EE to SQL 2005 SE.
Is there any other best way to upgrade Enterprise Edition
to Standard Edition on In-Place or any other method? Its is my Test sever.

You can install the 2005 server as a side-by-side install and transfer all your objects from 2000 server to 2005 server.

Yes, I second Dinakar's suggestion. Side-By-Side installation is good and recommended option against In-Place method.
