I will be creating a scheduled backup job in SQL 2000. I am just unsure of whether SQL has to be shutdown during the backup for the backup to be successful (worried about open files).

NO. You can still use and access the database while the backup is in operation

You cannot run a SQL Server backup while SQL Server is shutdown, and you cannot run a SQL Server job while SQL Server is shutdown.

A very good point

unless your schedule job is the OS's schedule job and you just want to copy the MDF & LDF to backup devices
